HLA haplotypes in primary sclerosing cholangitis patients of admixed and non ‐European ancestry
Primary sclerosing cholangitis (PSC) is strongly associated with several human leukocyte antigen (HLA) haplotypes. Due to extensive linkage disequilibrium and multiple polymorphic candidate genes in the HLA complex, identifying the alleles responsible for these associations has proven difficult. We aimed to evaluate whether studying populations of admixed or non‐European descent could help in defining the causative HLA alleles. When assessing haplotypes carrying HLA‐DRB1*13:01 (hypothesized to specifically increase the susceptibility to chronic cholangitis), we observed that every haplotype in the Scandinavian PSC popu...

Gene polymorphism and HLA ‐G expression in patients with childhood‐onset systemic lupus erythematosus: A pilot study
Human leukocyte antigen‐G (HLA‐G) presents inhibitory functions in immune cells and is located in a chromosomal region associated with systemic lupus erythematosus (SLE) susceptibility. Polymorphisms in 3′ untranslated region (3′UTR) of HLA‐G gene may influence protein expression. To date, no study analyzing HLA‐G polymorphism and expression in childhood‐onset systemic lupus erythematosus (cSLE) has been conducted. Therefore, we investigated the influence of HLA‐G 3′UTR polymorphisms in 50 cSLE patients and 144 healthy controls. For the expression analysis, the control group included 26 healthy individual...

Thirteen novel canine dog leukocyte antigen ‐88 alleles identified by sequence‐based typing
Major histocompatibility complex (MHC) genes in mammals include highly polymorphic class I and class II genes that are critical for donor‐recipient matching for transplantation. Dogs have served as an effective, directly translatable model for stem/progenitor cell transplantation. Previous analyses of MHC class I genes in dogs point to a single highly polymorphic gene, dog leukocyte antigen (DLA)‐88, as an important factor in the success or failure of hematopoietic stem cell transplants. Fifty‐nine DLA‐88 alleles have been identified and reported so far. Here, we extend this list by presenting 13 novel DLA‐88 all...

HLA ‐A*02:687, a novel allele identified by sequence‐based typing in cord blood from a Korean woman
Summary statement A*02:687 showed one nucleotide difference with A*02:01:01:01 resulting in an amino acid change. HLA‐A is the second most polymorphic HLA class I gene, with more than 3,900 alleles having been identified according to the IPD‐IMGT/HLA Database (Release 3.28.0, April 2017). 1 In particular, HLA‐A*02 is a very common allotype in the Korean population with a frequency of 28.50%.2 In the Korean population, 11 A*02 alleles have been reported, and A*02:01 is the most frequent.2–5 Here, we report a newly identified A*02:687, a variant of A*02:01:01:01 allele, that was detected in a cord blood unit donated ...

Impact of HLA ‐DRB1 allele polymorphisms on control of HIV infection in a Peruvian MSM cohort
Associations between HLA class II polymorphisms and HIV control were assessed in a Peruvian MSM cohort. Among 233 treatment naïve HIV+ individuals, DRB1*13:02 was linked to elevated viral loads (P = .044) while DRB1*12:01 showed significantly lower viral set points (P = .015) and restricted a dominant T cell response to HIV Gag p24 (P = .038). The present work contributes to a better knowledge of the Peruvian immunogenetics and supports the important role of HLA class II restricted T cells in HIV control. (Source: Tissue Antigens)

Pronase treatment improves flow cytometry crossmatching results
Flow cytometry crossmatching (FC‐XM) is the most sensitive cell‐based method for detecting donor‐specific antibodies in clinical organ transplantation. Unfortunately, background FC‐XM reactivity is elevated in assays with B lymphocytes—partly because of nonspecific immunoglobulin binding by Fc receptors and B‐cell surface immunoglobulins. To reduce the background reactivity in a B‐cell FC‐XM assay, we treated lymphocytes with pronase (1 mg/mL for 30 minutes). This treatment drastically reduced the presence of kappa light chains and Fc receptors (CD32b), while the concomitant decrease in CD19, CD20 and maj...

Identification of 400 novel alleles at the HLA ‐A, ‐B, ‐C, ‐DRB1, ‐DQB1 loci from China Marrow Donor Program
Four hundred novel human leukocyte antigen (HLA) alleles were identified in Chinese individuals: 100 HLA‐A alleles, 100 HLA‐B alleles, 101 HLA‐C alleles, 28 HLA‐DRB1 alleles and 71 HLA‐DQB1 alleles. Comparing novel alleles with their most homologous allele, we found 72.73% non‐synonymous nucleotide substitutions, 21.13% silent mutations, 3.90% nonsense mutations and 3.25% frameshift mutation. 352 (88%) of the 400 novel alleles are single nucleotide substitution variants when compared with their most homologous alleles and other novel alleles differ from their most similar allele by more than 1 nucleotide substi...
