Drug monitoring of sunitinib in patients with advanced solid tumors: a monocentric observational French study
This study aimed to determine a threshold value of plasma drug exposure associated with the occurrence of grade 3‐4 toxicity, then the potential impact of TDM on clinical decision. Consecutive outpatients treated with sunitinib were prospectively monitored between days 21‐28 of the first cycle, then monthly until disease progression. At each consultation, the composite AUCƬ,ss (sunitinib + active metabolite SU12662) was assayed. The decisions taken during each consultation were matched with AUCƬ,ss and compared to the decisional algorithm based on TDM. 105 cancer patients and 288 consultations were matched with the c...

Rationale and design of the CANARI study: a case –control study investigating the association between prostate cancer and 5‐alpha‐reductase inhibitors for symptomatic benign prostate hypertrophy by linking SNIIRAM and pathology laboratories in a specific region in France
Abstract Benign prostate hypertrophy (BPH) could be associated with low urinary symptoms requiring medical treatment: 5‐alpha‐reductase inhibitors (5‐ARI) or ɑ‐blockers. Two clinical trials investigating 5‐ARI use in prostate cancer (PCa) primary prevention highlighted a potential safety signal with an increased risk of high‐grade PCa. Later observational studies failed to show similar results but have some limits. This paper focuses on describing the protocol of the CANARI study and its feasibility, as regards the matching process of two pseudo‐anonymous databases. The study concerned patients living in the...

Atropinic (Anticholinergic) Burden in Antipsychotic ‐Treated Patients
This study showed high values of anticholinergic burden in patients receiving antipsychotics. Thus, considering the potential noxious clinical impact of atropinic properties on cognitive functions, an appropriate approach should be to reduce prescription of antipsychotics with a high anticholinergic burden but also coprescription of other frequently associated atropinic drugs, like antiparkinsonians, H1 antihistamines or imipraminic antidepressants in these patients. The impact of age and sex on the reporting of cough and angioedema with renin –angiotensin system inhibitors: a case/noncase study in VigiBase
Abstract The purpose of this study was to assess the impact of age and sex on the reporting of cough and angioedema related to renin–angiotensin system (RAS) inhibitors. A case/noncase study was performed in VigiBase. Two case groups were identified, reports of cough and reports of angioedema, and noncases were all reports of all other adverse events. Logistic regression analysis was used to assess the association between reporting of cough and angioedema with each class of RAS inhibitors stratified by age/sex and to control for confounding. The reporting of cough with angiotensin‐converting enzyme (ACE) inhibitors was...
