Genomewide screening for genes involved in biofilm formation and miconazole susceptibility in Saccharomyces cerevisiae
Abstract Infections related to fungal biofilms are difficult to treat due to the reduced susceptibility of sessile cells to most antifungal agents. Previous research has shown that 1–10% of sessile Candida cells survive treatment with high doses of miconazole (a fungicidal imidazole). The aim of this study was to identify genes involved in fungal biofilm formation and to unravel the mechanisms of resistance of these biofilms to miconazole. To this end, a screening of a Saccharomyces cerevisiae deletion mutant bank was carried out. Our results revealed that genes involved in peroxisomal transport and the biogenesis of the...

BH3‐only proteins Noxa, Bik, Bmf, and Bid activate Bax and Bak indirectly when studied in yeast model
Abstract BH3‐only proteins of the Bcl‐2 family regulate programmed cell death in mammals through activation of multidomain proapoptotic proteins Bax and Bak in response to various proapoptotic stimuli by mechanism that remains under dispute. Here, we report that the cell death‐promoting activity of BH3‐only proteins Bik, Bmf, Noxa, and tBid can only be reconstituted in yeast when both multidomain proapoptotic and antiapoptotic Bcl‐2 family proteins are present. Inability of these proteins to induce cell death in the absence of antiapoptotic proteins suggests that all tested BH3‐only proteins likely activate Bax...

Phenothiazine is a potent inhibitor of prostaglandin E2 production by Candida albicans biofilms
This study investigated the effect of PTZ on biofilm formation, ABC transporters and PGE2 production by C. albicans. This was done by growing C. albicans biofilms in the presence of AA and PTZ and measuring the biomass as well as reduction of 2,3‐bis(2‐methoxy‐4‐nitro‐5‐sulfophenyl)‐5[(phenylamino) carbonyl]‐2H tetrazolium hydroxide. The effect on ABC transporters was determined by rhodamine 6G efflux and the concentration PGE2 was determined by a monoclonal PGE2 enzyme‐linked immunosorbent assay and LC/MS/MS. Our results indicate that PTZ can cause a reduction in both the metabolic activity and biomass o...
