Understanding the antibody repertoire in Neuropsychiatric Systemic Lupus Erythematosus and Neuromyelitis Optica Spectrum Disorders: do they share common targets?
ConclusionPatients with demyelinating disease should be tested for AQP4‐IgG, MOG‐IgG and DWEYS‐IgG. AQP4–IgG is diagnostic for NMOSD; none of the antibodies is diagnostic for dNPSLE. dsDNA‐IgG are present in NMOSD but are not cross‐reactive with DWEYS, indicating a different antigenic stimulus and potentially different mechanisms of tissue damage.This article is protected by copyright. Antigen ‐driven lymphoproliferations in Sjögren's Syndrome patients accumulate in complement receptor 2/CD21‐/low B cells
ConclusionClonal lymphoproliferations in SS patients preferentially accumulate in the autoreactive CD21‐/low B cell compartment, which is often amplified in these subjects, and (self)‐antigen recognition may drive expansion while further refining BCR (self)‐reactivity.This article is protected by copyright. Belimumab for the Treatment of Early Diffuse Systemic Sclerosis: Results of a Randomized, Double ‐Blind, Placebo‐Controlled, Pilot Trial
ConclusionPatients in both treatment groups experienced significant improvements in MRSS. The median difference was greater in the belimumab group but did not achieve statistical significance in this small pilot study. Adverse events were similar between the groups. Changes in gene expression were consistent with mechanism of action and show that clinical response to treatment with belimumab is associated with significant decrease in pro‐fibrotic genes and pathways. Gut Microbiota Perturbations in Reactive Arthritis and Post ‐Infectious Spondyloarthritis
ConclusionThis is the first culture‐independent study characterizing the gut microbial community of post‐infectious arthritis. Although bacterial factors correlated with disease presence and clinical features of ReA, host genetics also appeared to be a major independent driver of intestinal community composition. Understanding of these gut microbiota host‐genetic relationships may further clarify the pathogenesis of post‐infectious spondyloarthropathies.This article is protected by copyright. Brief Report: Association of Natural Killer Cell Ligand Polymorphism HLA –C Asn80Lys With the Development of Anti‐SSA/Ro–Associated Congenital Heart Block
ConclusionThese data establish C2 as a novel genetic risk factor associated with CHB. This observation supports a model in which fetuses with C2 ligand expression and maternal anti‐SSA/Ro positivity may have impaired NK cell surveillance, resulting in unchecked cardiac inflammation and scarring. Role of Stromelysin 2 (Matrix Metalloproteinase 10) as a Novel Mediator of Vascular Remodeling Underlying Pulmonary Hypertension Associated With Systemic Sclerosis
ConclusionGene expression profiling of EPC‐derived ECs identified MMP10 as a novel candidate gene in SSc‐associated PH. MMP10 is overexpressed in the serum and pulmonary arteries of patients with SSc‐associated PH, and its blockade alleviates PH in the Fra‐2–Tg mouse model. NF ‐κB2 Controls the Migratory Activity of Memory T Cells by Regulating Expression of CXCR4 in a Mouse Model of Sjögren's Syndrome
ConclusionOur results suggest that the RelB/NF‐κB2 pathway regulates T cell migration to autoimmune targets through TGFβ/TGFβR‐dependent regulation of CXCL12/CXCR4 signaling.
