Long-term decomposition of aqueous S-nitrosoglutathione and S-nitroso-N-acetylcysteine: Influence of concentration, temperature, pH and light
Publication date: Available online 7 January 2019Source: Nitric OxideAuthor(s): Gabriela Freitas P. de Souza, Jaqueline Priscilla Denadai, Guilherme F. Picheth, Marcelo Ganzarolli de OliveiraAbstractPrimary S-nitrosothiols (RSNOs) have received significant attention for their ability to modulate NO signaling in many physiological and pathophysiological processes. Such actions and their potential pharmaceutical uses demand a better knowledge of their stability in aqueous solutions. Herein, we investigated the effects of concentration, temperature, pH, room light and metal ions on the long-term kinetic behavior of two repres...

S-1-Propenylcysteine augments BACH1 degradation and heme oxygenase 1 expression in a nitric oxide-dependent manner in endothelial cells
Publication date: Available online 8 January 2019Source: Nitric OxideAuthor(s): Tadamitsu Tsuneyoshi, Kayo Kunimura, Naoaki MoriharaAbstractGarlic has been demonstrated to exert protective effects against oxidative damage using numerous experimental models. The antioxidant effects of garlic are associated with the activation of Nrf2-dependent gene expression. S-1-Propenylcysteine (S1PC) and S-allylcysteine (SAC) are two predominant sulfur amino acids present in aged garlic extract; however, the exact roles of these amino acids within the Keap1/Nrf2 system remain unknown. We hypothesized that sulfur-containing amino acids d...

Nitric oxide generation from S-nitrosoglutathione: New activity of indium and a survey of metal ion effects
In this study, the ability of various transition and post-transition metal ions to promote NO release from GSNO was surveyed by chemiluminescence-based NO detection. Substantial NO formation (>10-fold increase relative to GSNO baseline) was detected after the addition of Cu2+, Au3+, Pd2+, Pt2+, and V3+. Modest increases were observed in the cases of Co2+, Hf4+, Fe2+, Fe3+, Mn2+, Hg2+, Ni2+, Ag+, Sn2+, and Zr4+, while no effect was evident for Al3+, Cr3+, Pb2+, Sc3+, and Zn2+. It was further observed that In+ compounds initiate the apparent NO-forming decomposition of GSNO, while In0 and In3+ are inactive, indicating that I...

Hydrogen peroxide and nitric oxide induce anticontractile effect of perivascular adipose tissue via renin angiotensin system activation
ConclusionFor the first time in the literature, our results show the presence of Mas and AT2 receptors, as well as, nitric oxide synthase on perivascular adipose tissue. Furthermore, our results show the involvement of Mas and AT2 receptors and consequently nitric oxide synthase activation in the anti-contractile effect exerted by perivascular adipose tissue.Graphical abstractRepresentative figure of the PVAT vasodilator pathway. Mas or AT2 receptors activation in PVAT culminates in the PI3K – Akt pathway and NOS activation that induce the production NO and H2O2 from eNOS and nNOS. (Source: Nitric Oxide)

Antioxidant activity of endogenously produced nitric oxide against the zinc oxide nanoparticle-induced oxidative stress in primary hepatocytes of air-breathing catfish, Clarias magur
Publication date: Available online 31 December 2018Source: Nitric OxideAuthor(s): Debaprasad Koner, Bodhisattwa Banerjee, Rubaiya Hasan, Nirmalendu SahaAbstractThe facultative air-breathing magur catfish (Clarias magur) regularly encounter various environmental challenges including the exposure to nanomaterials discarded as industrial wastes in water bodies. The present investigation aimed at determining the possible ZnO NP-induced oxidative stress and also the antioxidant strategy of nitric oxide (NO), generated endogenously, in primary hepatocytes of magur catfish. Exposure of primary hepatocytes to different concentrati...

Differential expression of alternative transcripts of soluble guanylyl cyclase, GYCY1a3 and GUCY1b3 genes, in the malignant and benign breast tumors
Publication date: Available online 28 December 2018Source: Nitric OxideAuthor(s): Milad Mohammadoo khorasani, Fatemeh Karami Tehrani, Seyed Mohammad Reza Parizadeh, Morteza AtriAbstractExtensive alterations in splicing is one of the molecular indicator for human cancers. Soluble guanylyl cyclase (sGC), an obligatory heterodimer, is composed of α1 and β1 subunits. Each subunit is encoded by a separate gene, GUCY1a3 and GUCY1b3, correspondingly. sGC activity has been regulated by an alternative splicing and it has an important effect on the breast cancer. sGC alternative splicing has been evaluated in the 55 malignant, 25 ...

Regulation of endothelial barrier integrity by redox-dependent nitric oxide signaling: Implication in traumatic and inflammatory brain injuries
Publication date: Available online 24 December 2018Source: Nitric OxideAuthor(s): Seungho Choi, Nishant Saxena, Tajinder Dhammu, Mushfiquddin Khan, Avtar K. Singh, Inderjit Singh, Jeseong WonAbstractNitric oxide (NO) synthesized by eNOS plays a key role in regulation of endothelial barrier integrity but underlying cell signaling pathway is not fully understood at present. Here, we report opposing roles of two different redox-dependent NO metabolites; peroxynitrite (ONOOˉ) vs. S-nitrosoglutathione (GSNO), in cell signaling pathways for endothelial barrier disruption. In cultured human brain microvessel endothelial cells (h...

Nitric oxide synthase inhibitors 1400W and L-NIO inhibit angiogenesis pathway of colorectal cancer
ConclusionNOS inhibitors could suppress colorectal cancer cell growth and migration, likely via suppressing the angiogenesis pathway.Graphical abstractNO is implicated in angiogenesis, a hallmark for CRC. By inhibiting NOS using 1400W or L-NIO, CRC cell growth and migration were suppressed, with simultaneous inhibition of the angiogenic pathway. (Source: Nitric Oxide)

Mikoyan, Svetlana L. Kotova, Tatyana G. Rudenko, Alexey Fayzullin, Peter S. TimashevAbstractWe have studied the effect of interactions between dinitrosyl iron complexes with thiol-containing ligands (DNIC-TL) and diglucamine salt of chlorine e6 (photoditazine, PD) on the rate of photosensitized oxidation of a model organic substrate – tryptophan – in the presence and absence of an amphiphilic polymer, Pluronic F127, as well as on ...

Nitric oxide affects cadmium-induced changes in the lichen Ramalina farinacea
Publication date: Available online 7 December 2018Source: Nitric OxideAuthor(s): Jozef Kováčik, Sławomir Dresler, Giuseppe Micalizzi, Petr Babula, Juraj Hladký, Luigi MondelloAbstractMetabolic responses of epiphytic lichen Ramalina farinacea to cadmium (Cd) and/or nitric oxide (NO) scavenger (cPTIO) were studied. Accumulation of Cd and other metallic nutrients was not affected by cPTIO while total and absorbed amounts differed. Cd-induced NO formation was suppressed by cPTIO but ROS signal was synergistically enhanced, confirming that NO is essential to keep ROS under control. This excessive ROS generation could be a r...
