Nitric oxide releasing two-part creams containing S-nitrosoglutathione and zinc oxide for potential topical antimicrobial applications
In this study, the potential for creating NO releasing creams containing the naturally occurring NO carrier, S-nitrosoglutathione (GSNO), are characterized and evaluated. GSNO is shown to have prolonged stability (>300 days) when mixed and stored within Vaseline at 24 °C. Further, enhanced proliferation of NO from GSNO using zinc oxide nanoparticles (ZnO) is demonstrated. Triggering NO release from the GSNO/Vaseline mixture using a commercial zinc oxide-containing cream exhibits first-order NO release kinetics with the highest %NO release over the first 6 h. Significant killing effects against S. aureus, S. epidermidi...

New generation of nitric oxide-releasing porous materials: Assessment of their potential to regulate biological functions
Publication date: Available online 30 May 2019Source: Nitric OxideAuthor(s): Rosana V. Pinto, Ana C. Fernandes, Fernando Antunes, Zhi Lin, João Rocha, João Pires, Moisés L. PintoAbstractNitric oxide (NO) presents innumerable biological roles, and its exogenous supplementation for therapeutic purposes has become a necessity. Some nanoporous materials proved to be potential vehicles for NO with high storage capacity. However, there is still a lack of information about their efficiency to release controlled NO and if they are biocompatible and biologically stable. In this work, we address this knowledge gap starting by eva...

Hydrogen sulfide suppresses homocysteine-induced glial activation and inflammatory response
Publication date: Available online 28 May 2019Source: Nitric OxideAuthor(s): Mohit Kumar, Rajat SandhirAbstractNeuro-inflammation plays a critical role in hyperhomocysteinemia (HHcy)-associated neurodegenerative disorders. Hydrogen sulfide (H2S) has been suggested as an endogenous neuromodulator and potent anti-inflammatory molecule. In present study, we have investigated the effect of NaHS supplementation (a H2S source) on inflammatory response in animals subjected to HHcy. NaHS adminstration restored the decreased levels of H2S and polysulfides with a concomitant increase in the activity of cystathionase (CSE) and cystat...

Light-induced release of nitric oxide from the nitric oxide-bound CDGSH-type [2Fe–2S] clusters in mitochondrial protein Miner2
Publication date: Available online 28 May 2019Source: Nitric OxideAuthor(s): Yiming Wang, Jeonghoon Lee, Huangen DingAbstractHuman mitochondrial matrix protein Miner2 hosts two [2Fe–2S] clusters via two CDGSH (Cys-Asp-Gly-Ser-His) motifs. Unlike other iron-sulfur clusters in proteins, the reduced CDGSH-type [2Fe–2S] clusters in Miner2 are able to bind nitric oxide (NO) and form stable NO-bound [2Fe–2S] clusters without disruption of the clusters. Here we report that the NO-bound Miner2 [2Fe–2S] clusters can quickly release NO upon the visible light excitation. The UV–visible and Electron Paramagnetic Resonance (E...

Nitric oxide and abscisic acid protects against PEG-induced drought stress differentially in Brassica genotypes by combining the role of stress modulators, markers and antioxidants
Publication date: Available online 13 May 2019Source: Nitric OxideAuthor(s): Seema Sahay, Ehasanullah Khan, Meetu GuptaAbstractThe present study was designed to see the effect of exogenous nitric oxide (NO) and abscisic acid (ABA) and their interaction on physiological and biochemical activities in leaves and roots of two Indian mustard (Brassica juncea) cultivars [cv. Pusa Jagannath (PJN) and Varuna (VAR)] exposed to polyethylene glycol (PEG)-induced drought stress. Seven days old hydroponically grown seedlings were treated with PEG (10%), sodium nitroprusside, a NO donor [NO (100 μM)] and abscisic acid [ABA (10 μM)],...

Exogenous l-ARGININE does not stimulate production OF NO or cGMP within the rat corporal smooth muscle cells in culture
ConclusionsThis in vitro study demonstrates that exogenous administration of L-ARG or L-CIT failed to stimulate production of either NO or cGMP by the corporal CSM cells. A re-evaluation of the presumptive role of the exogenous administration of L-ARG in improving the synthesis of NO at least at the level of the CSM cells appears warranted. (Source: Nitric Oxide)

NADPH oxidase is a primary target for antioxidant effects by inorganic nitrite in lipopolysaccharide-induced oxidative stress in mice and in macrophage cells
In conclusion, these novel findings position NADPH oxidase in the inflammatory vasculature as a prime target for the antioxidant effects of inorganic nitrite, and open a new direction to modulate the inflammatory response.Graphical abstractThrough mediating NADPH oxidase activity, the increase of NO bioavailability by nitrite-NO pathway will reduce vascular oxidative stress and endothelial dysfunction, resulting in higher survival during acute inflammation. The role of nitric oxide signaling in pulmonary circulation of high- and low-altitude newborn sheep under basal and acute hypoxic conditions
Publication date: Available online 4 May 2019Source: Nitric OxideAuthor(s): Emilio A. Herrera, Germán Ebensperger, Ismael Hernández, Emilia M. Sanhueza, Aníbal J. Llanos, Roberto V. ReyesAbstractNitric oxide (NO) is the main vasodilator agent that drives the rapid decrease of pulmonary vascular resistance for the respiratory onset during the fetal to neonatal transition. Nevertheless, the enhanced NO generation by the neonatal pulmonary arterial endothelium does not prevent development of hypoxic pulmonary hypertension in species without an evolutionary story at high altitude. Therefore, this study aims to describe the ...

Alveolar concentration of nitric oxide as a prognostic biomarker in idiopathic pulmonary fibrosis
ConclusionWe confirm that CaNO was significantly higher in IPF patients than in healthy controls and its correlation with functional parameters. Moreover, CaNO ≥6 and ≥9 ppb were significantly correlated with mortality and disease progression, respectively. These data suggest that CaNO, a non-invasive and reproducible biomarker, may predict disease progression and survival outcome in patients with IPF. (Source: Nitric Oxide)

Dietary nitrate supplementation alters the oral microbiome but does not improve the vascular responses to an acute nitrate dose
Publication date: Available online 30 April 2019Source: Nitric OxideAuthor(s): Mia Burleigh, Luke Liddle, David J. Muggeridge, Christopher Monaghan, Nicholas Sculthorpe, John Butcher, Fiona Henriquez, Chris EastonAbstractNitrate (NO3−) contained in food and beverages can transiently increase nitric oxide (NO) availability following a stepwise reduction to nitrite (NO2−) by commensal bacteria in the oral cavity. We tested the hypothesis that regular ingestion of dietary NO3− would influence the oral microbiome, the capacity to reduce NO3− to NO2− in saliva, and the vascular responses to an acute dose of NO3−. Th...
