Role of sialic acid ‐containing glycans of matrix metalloproteinase‐9 (MMP‐9) in the interaction between MMP‐9 and staphylococcal superantigen‐like protein 5
ABSTRACT Staphylococcal superantigen‐like proteins (SSL) show no superantigenic activity but have recently been considered to act as immune suppressors. It was previously reported that SSL5 bound to P‐selectin glycoprotein ligand‐1 (PSGL‐1) and matrix metalloproteinase (MMP)‐9, leading to inhibition of leukocyte adhesion and invasion. These interactions were suggested to depend on sialic acid‐containing glycans of MMP‐9, but the roles of sialic acids in the interaction between SSL5 and MMP‐9 are still controversial. In the present study, we prepared recombinant glutathione S‐transferase‐tagged SSL5 (GST...

The immune characterization of IFN ‐β responses in tuberculosis patients
Abstract We aimed to assess the immunoregulatory effects of IFN‐β in patients with tuberculous pleurisy. IFN‐β, IFN‐γ and IL‐17 expression levels were detected, and correlations among these factors in different culture groups were analyzed. Pleural fluid mononuclear cells (PFMCs) from tuberculous pleural effusions, but not peripheral blood mononuclear cells (PBMCs) from healthy donors, spontaneously expressed IFN‐β, IL‐17 and IFN‐γ. Moreover, exogenous IFN‐β significantly inhibited the expression of IL‐17 in PFMCs. By contrast, IFN‐β simultaneously enhanced the levels of IFN‐γ. To further inve...

Characterization of morphological conversions of Helicobacter pylori under anaerobic conditions
In this study, we used scanning electron microscopy to document the anaerobiosis‐induced morphological change of H. pylori, from helical to coccoid to the newly defined fragmented form. Western blot analysis indicated that all three forms expressed certain pathogenic proteins, including the bacterial cytotoxin‐associated gene A (CagA), components of the cag‐Type IV secretion system (TFSS), the blood group antigen‐binding adhesin BabA, and UreA (apoenzyme of urease) at nearly equivalent levels. Similar urease activities were also detected in each form of H. pylori. However, in contrast to the helical form, the anaer...

Total HIV DNA decreases more pronouncedly in patients infected by CRF01_AE than subtype B and CRF07_BC after 18 months of antiretroviral therapy
ABSTRACT Whether the amount of HIV DNA was associated with the various subtype of HIV‐1 after antiretroviral therapy (ART) has not been reported. In the present study, the amount of HIV DNA and RNA as well as the CD4+T counts in blood and semen prior to and after 18 months of ART were compared in 48 patients infected by CRF01_AE, subtype B or CRF07_BC of HIV‐1. All the patients achieved successful viral RNA suppressions and CD4 cell count recovery. The total HIV DNA levels were similar before ART, however the lowest level of HIV DNA was observed in patients with CRF01_AE after ART. According to co‐receptor usage pred...

Bifidobacteria alleviate experimentally induced colitis by upregulating indoleamine 2, 3 ‐dioxygenase expression
ABSTRACT The goal of this study was explore the role of indoleamine 2, 3‐dioxygenase (IDO) in the therapeutic effect of probiotics on inflammatory bowel disease (IBD). Trinitrobenzene sulfonic acid (TNBS) was used to induce colitis in mice and 1‐methyltryptophan (1‐MT) to block expression of IDO. Clinical manifestations and macroscopic and microscopic colonic changes were assessed using a disease activity index (DAI), the Wallace–Keenan, and Curtner scoring systems, respectively. Expression of colonic IDO was detected by western blot. Immunohistochemistry analysis to evaluate numbers of CD11c+ cells and expression ...

The 1st step initiation essential for allergen ‐specific IgE antibody production upon the 2nd step: Induction of non‐specific IgE+ small B cells containing secondly‐sensitized allergen‐specific ones in mice firstly‐sensitized with an allergen
ABSTRACT There was a significant amount of non‐specific, but not of allergen (e.g., papain, mite feces and four kinds of pollen)‐specific, IgE antibodies (Abs) in the sera of normal mice. An i.n. injection of each allergen without adjuvant into mice caused an increase in total IgE Ab titers with a similar time course in the serum. However, the stage of initiation of allergy varied from allergen to allergen. Submandibular lymph node cells from normal mice contained papain‐, but not mite feces‐ or pollen‐specific IgE+ cells and an i.n. injection of papain induced papain‐specific IgE Abs in the serum. In contrast,...
