Analysis of tauopathy research funding between 2006 and 2016 reveals critical gaps in research priorities
Publication date: Available online 9 October 2018Source: Alzheimer's & DementiaAuthor(s): Cara M. Altimus, Kirstie Keller, Patrick Brannelly, Erin Ross, Chang-Ting Lin, Ekemini A.U. Riley, LaTese Briggs, Jeremy Smith, Melissa StevensAbstractNeurodegenerative diseases encompass a range of diagnoses, such as Alzheimer's disease and Parkinson's disease. Despite decades of advancements in understanding the neurobiology of individual diseases, this class has few disease-modifying therapeutics and a paucity of biomarkers for diagnosis or progression. However, tau protein aggregation has emerged as a potential unifying factor acr...

Participant satisfaction with dementia prevention research: Results from Home-Based Assessment trial
DiscussionOlder adults' participation in research was primarily motivated by altruism. Methodologies that facilitate human contact, encourage feedback and novelty of tasks should be incorporated into future trial design. (Source: Alzheimer's and Dementia: The Journal of the Alzheimer's Association)

Genetic overlap between vascular pathologies and Alzheimer's dementia and potential causal mechanisms
DiscussionOur findings provide evidence for genetic overlap, mostly due to apolipoprotein E (APOE), between vascular pathologies and AD dementia. The association between AD polygenic risk and late-life cognition is mediated in part via effects on vascular pathologies. An updated Alzheimer hypothesis: Complement C3 and risk of Alzheimer's disease—A cohort study of 95,442 individuals
DiscussionLow baseline levels of complement C3 were associated with high risk of Alzheimer's disease. The risk was amplified in APOE ε44 highly susceptible individuals, and these findings were substantiated by a Mendelian randomization approach, potentially implying causality. Based on these findings, we present and thoroughly discuss an updated Alzheimer hypothesis incorporating low complement C3 levels. Progress and future challenges in aging and diversity research in the United States
This report disseminates the research priorities that emerged from this conference, building on an earlier Gerontological Society of America preconference. We review key racial/ethnic differences in cognitive aging and dementia and identify current knowledge gaps in the field. We advocate for a systems-level framework for future research whereby environmental, sociocultural, behavioral, neuropathological, genetic, and psychometric levels of analysis are examined together to identify pathways and mechanisms that influence disparities. We then discuss steps to increase the recruitment and retention of racial/ethnic minoritie...
