1, 25-dihydroxy vitamin D3 inhibits Ras-MEK-ERK pathway and regulates proliferation and apoptosis of papillary thyroid carcinoma
Conclusion1, 25-(OH)2D3 can inhibit the activity and promote apoptosis of papillary thyroid carcinoma cell line TPC-1, and its mechanism may be related to the inhibition of Ras-MEK-ERK pathway activity, thus affecting the proliferation and the expression of apoptosis-related proteins. Daturmetesides A-E, five new ergostane-type C28 sterols from the leaves of Datura metel L
Publication date: Available online 23 January 2020Source: SteroidsAuthor(s): Jin-Yan Tan, Yan Liu, Yan-Gang Cheng, Yan-Ping Sun, Yuan Liu, Jin Huang, Shuang Guo, Guo-Zhen Liu, Hai-Xue Kuang, Bing-You YangAbstractFive undescribed ergostane-type C28 sterols, daturmetesides A-E (1-5), were isolated from the leaves of Datura metel L. The chemical structures of these new compounds were characterized through extensive spectroscopic analysis and comparison with literatures. Among them, the absolute structures of daturmetesides A and C were unambiguously determined by X-ray crystallography. The anti-inflammatory effect of daturmet...

Steroidal saponins from Trillium tschonoskii rhizomes and their cytotoxicity against HepG2 cells
Publication date: Available online 23 January 2020Source: SteroidsAuthor(s): Yin-Jun Yang, Xu Pang, Bei Wang, Jie Yang, Xiao-Juan Chen, Xin-Guang Sun, Qi Li, Jie Zhang, Bao-Lin Guo, Bai-ping MaAbstractA phytochemical study on the rhizomes of Trillium tschonoskii led to the isolation of fourteen new steroidal saponins, trillitschosides S1-S14 (1–14), along with ten known analogues (15-24). Their structures were established mainly by spectroscopic analyses as well as necessary chemical evidence. All isolated compounds were screened for the cytotoxicity against HepG2 cells, and the results demonstrated that only the known c...

Molecular structure study of a heptakis(2,3,6-tri-O-methyl)-β-cyclodextrin complex of cholesterol
Publication date: March 2020Source: Steroids, Volume 155Author(s): Karolína Kučáková, Bohumil DolenskýAbstractHeptakis(2,3,6-tri-O-methyl)-β-cyclodextrin (2) and cholesterol form a water-soluble complex 3. We performed several NMR studies, particularly 1H, 13C, 2D NOESY and DOSY, at various temperatures on 500 and 950 MHz instruments. We discovered that the complex 3 is unstable above 57 °C in heavy water, while it is kinetically stable enough to be studied by NMR in detail at 1 °C. We demonstrated the formation of a face-to-face 2:1 complex with a binding constant of approximately 2.2 × 106 M−2.Graphical a...

25-Hydroxyvitamin D3 positively regulates periodontal inflammaging via SOCS3/STAT signaling in diabetic mice
ConclusionDiabetic periodontitis appeared to develop an inflammaging status resulted in periodontal infection. 25(OH)D3 could inhibit SASP secretion through reducing SOCS3 expression in experimental diabetic periodontitis, dependently inactivating NF-κB pro-inflammatory signaling. The reversible effect further documented that the inflammaging might be a highly likely contributor in diabetic periodontitis.Graphical abstract25(OH)D3 played the protective role in gingival tissue of db/db mice with periodontitis. SOCS3 is an endogenous negative regulator of the inflammaging. Diabetic periodontitis developed an inflammaging st...

Training counteracts DEX-induced microvascular rarefaction by improving the balance between apoptotic and angiogenic proteins
Publication date: Available online 2 January 2020Source: SteroidsAuthor(s): Isley de Jesus, Naiara A. Herrera, Jesus C. Andreo, Carlos F. Santos, Sandra L. AmaralAbstractThis work investigated the mechanisms induced by exercise training that may contribute to attenuate dexamethasone (DEX)-induced microvascular rarefaction and hypertension. Wistar rats underwent training protocol or were kept sedentary for 8 weeks. Dexamethasone was administered during the following 14-days and hemodynamic parameters were recorded at the end. Capillary density (CD) and capillary-to-fiber ratio (C:F ratio) were obtained in soleus muscle (SOL...

Is 9β-dehydrohalogenation of betamethasone and dexamethasone hindering the detection of banned co-eluting meprednisone? A reverse-phase chiral liquid chromatography high-resolution mass spectrometry approach
Publication date: Available online 3 January 2020Source: SteroidsAuthor(s): Tajudheen K. Karatt, M. Anwar Sathiq, Saraswathy LayaAbstractMass spectral analyses of dexamethasone and betamethasone reveal intense signals at m/z 373.19994 (using a Thermo Q Exactive high-resolution mass spectrometer coupled with Dionex UltiMate 3000 UHPLC+ operated in the positive ion mode), matching the signal of meprednisone, the 11-oxo version of methylprednisolone, along with its parent signal; possibly due to dehydrohalogenation of these drugs at MS. The parent mass of meprednisone is exactly same as that of dehydrohalogenated mass of dexa...

Sexual dimorphism in the microbiology of the CF ‘Gender Gap’: Estrogen modulation of Pseudomonas aeruginosa virulence
In conclusion, we have demonstrated for the first time that estrogen exacerbates P. aeruginosa virulence factors and enhances bacterial interactions with CF bronchial epithelium which can be inhibited by tamoxifen. Our work suggests that SERMs could be used as an adjuvant treatment to block estrogen-induced P. aeruginosa infections and associated lung exacerbations in females with CF.Graphical abstract (Source: Steroids)
