Host‐cell specificity and transcytosis of nontoxic nonhemagglutinin protein of botulinum neurotoxin serotype D
Abstract Serotype D botulinum toxin (BoNT) complex (TC), a causative agent of foodborne botulism in animals, traverses the gastrointestinal tract and circulation, eventually becoming localized in neuromuscular junctions, where the serotype D BoNT cleaves SNARE substrate synaptobrevin II involved in neurotransmitter release. During this process, BoNT must pass through cells, thus from the intestinal lumen to the cells of the intestinal tract and blood vessels. The botulinum TC is formed by association of the BoNT with at least one nontoxic protein, which may be a nontoxic nonhemagglutinin (NTNHA). In this work, we examined ...

Intracellular glycerol accumulation in light‐limited Dunaliella tertiolecta culture is determined by partitioning of glycerol across the cell membrane
Abstract Dunaliella accumulates intracellular glycerol to counterbalance the extracellular salinity. In N‐limited chemostat cultures of D. tertiolecta, total glycerol production (sum of intracellular and extracellular) and intracellular glycerol content were proportional to the salinity of the culture medium. In the light‐limited D. tertiolecta culture, total glycerol output (sum of intracellular and extracellular) was relatively constant at different salinities (0.5 and 2.0 M), while the intracellular glycerol content was proportional to the culture medium salinity, that is, the cells released less glycerol into th...

Incubation temperature, osmolarity, and salicylate affect the expression of resistance–nodulation–division efflux pumps and outer membrane porins in Acinetobacter baumannii ATCC19606T
In this study, we examined the impact of various environmental conditions on the expression of resistance–nodulation–division (RND) efflux pumps and outer membrane (OM) porins, two key determinants of Acinetobacter baumannii's intrinsic resistance, an organism known to cause various multidrug resistant infections in immunocompromised individuals. Quantitative RT‐PCR was used to analyze the expression of adeB, adeG, and adeJ (genes encoding RND pumps) and 33 kDa, carO, and oprD (genes encoding OM porins) of A. baumannii ATCC19606T under different incubation temperatures (30, 37, and 42 °C) and in the presence of h...

The effect of specific rhlA‐las‐box mutations on DNA binding and gene activation by Pseudomonas aeruginosa quorum‐sensing transcriptional regulators RhlR and LasR
Abstract Pseudomonas aeruginosa is a free‐living bacterium and an important opportunistic pathogen. The genes coding for virulence‐associated traits are regulated at the level of transcription by the quorum‐sensing response. In this response, the regulator LasR coupled with the autoinducer 3‐oxo‐dodecanoyl homoserine lactone (3O‐C12‐HSL) activates transcription of genes for several virulence factors. LasR/3O‐C12‐HSL also activates transcription of rhlR, the gene coding for the transcriptional regulator RhlR, and of rhlI that encodes the synthase that produces the autoinducer butanoyl‐homoserine lactone ...
