Controlling Anthracnose By Means of extracts, and their major constituents, from Brosimum rubescens Taub
Publication date: Available online 30 November 2019Source: Biotechnology ReportsAuthor(s): Janio Martinez, Andrés Gomez, Cesar Ramirez, Jesús Gil, Diego DurangoAbstractAnthracnose, caused by the fungus Colletotrichum gloeosporioides (Penz.) Penz. & Sacc., is the most limiting fungal disease of mango and papaya crops in Colombia. The in vivo and in vitro activity against C. gloeosporioides of the extracts from sawdust of Brosimum rubescens Taub. (Moraceae) was evaluated. The extracts of less polarity (n-hexane and dichloromethane) displayed the greatest inhibitory effects. Then, the coumarins xanthyletin (2.74% d.w.) and ...

Selective and sensitive colorimetric detection of platinum using Pseudomonas stutzeri mediated optimally synthesized antibacterial silver nanoparticles
Publication date: Available online 22 November 2019Source: Biotechnology ReportsAuthor(s): Megha P. Desai, Reshma V. Patil, Kiran D. PawarAbstractIn this work, Pseudomonas stutzeri was used for the optimum biogenic synthesis of antibacterial silver nanoparticles (AgNPs) which were applied for colorimetric detection of platinum ions (Pt+2). The optimum synthesis conditions were 2 mM AgNO3, pH 9 and incubation at 60 °C for 24 h. The FTIR spectra indicated that biomolecules such as amino acids, proteins or enzymes from P. stutzeri were involved in the synthesis of AgNPs in the size range of 10-50 nm. Among the variou...

Draft genomes and initial characteriaztion of siderophore producing pseudomonads isolated from mine dump and mine drainage
Publication date: Available online 23 November 2019Source: Biotechnology ReportsAuthor(s): Marika Hofmann, Thomas Heine, Vivian Schulz, Sarah Hofmann, Dirk TischlerAbstractSiderophores are of high interest for biotechnological, pharmaceutical, agricultural and industrial applications. Although they are synthesized by various organisms, the yield is usually low which hindrances their suitability for broad range uses. Thus, it is necessary to identify novel producers and to increase the understanding of the biosynthesis pathways. Herein we report the isolation of two novel Pseudomonas strains and the identification of the ge...

Coproduction of lipids and extracellular polysaccharides from the novel green alga Parachlorella sp. BX1.5 depending on cultivation conditions
Publication date: Available online 21 November 2019Source: Biotechnology ReportsAuthor(s): Mitsuki Sasaki, Akari Takagi, Shuhei Ota, Shigeyuki Kawano, Daisaku Sasaki, Munehiko AsayamaAbstractA novel strain of microalga Parachlorella sp. BX1.5 was isolated and its unique properties of producing lipids and extracellular polysaccharides (EPS) characterized. The cells could extracellularly produce a large amount of acidic EPS, when cultured in nitrogen-deficient BG110 medium (BG11—N) with 2% CO2-air supply. The main component of intracellularly accumulated lipids was triacylglycerol (TAG), depending on the different cultivat...

Biogas production from recycled paper mill wastewater by UASB digester: optimal and mesophilic conditions
In this study, the AD experiment was performed in continuous mode with an effluent inlet flow rate equal to 1 L/h. Using the optimal OLR value 5.18 g COD/Ld and with an effluent 5,7 g COD/L, a biogas yield of 92 N mL/g COD removed (at normal temperature and pressure) was obtained during the RPMW anaerobic treatment. The reactor was operated at an optimal hydraulic retention time (HRT) of 15.14 h with a biogas production volume of the optimal value 62.5 L/d. These results indicate that RPMW can be effectively treated in a UASB reactor with the advantage of producing biogas. We tested our system with RPMW, to s...

Household aluminum foil matte and bright side reflectivity measurements: Application to a photobioreactor light concentrator design
Publication date: Available online 20 November 2019Source: Biotechnology ReportsAuthor(s): Victor Pozzobon, Wendie Levasseur, Khanh-Van Do, Bruno Palpant, Patrick PerréAbstractThis work reports the design of a light concentrator intended to be used to cast uniform lighting over a photobioreactor. Household aluminum foils was chosen as reflective material to build the concentrator. This choice raised the question of which side to use. Thus measurements of household aluminum foil reflectivity spectra on both bright and matte sides were undergone. These measurements were done using an integrating sphere, over a 250–2500&nb...

Monitoring method for transgene expression in target tissue by blood sampling
In this study, we have developed a novel method to monitor transgene expression in tissues by blood sampling. We administered plasmid DNA (pDNA) encoding non-secretory form of firefly luciferase as a reporter gene and pDNA encoding secretable Gaussia princeps luciferase as a monitor gene simultaneously into mice. Good positive correlations were found between log-transgene expression of the reporter gene and the monitor gene in the treated muscle, between the monitor gene in the treated muscle and plasma, and consequently between the reporter gene in the treated muscle and the monitor gene in plasma after naked pDNA transfe...

Hyper-production of Taxol from Aspergillus fumigatus, an endophytic fungus isolated from Taxus sp. of the Northern Himalayan region
Publication date: Available online 15 November 2019Source: Biotechnology ReportsAuthor(s): Pradeep Kumar, Balwant Singh, Vikram Thakur, Abhishek Thakur, Nandita Thakur, Deepak Pandey, Duni ChandAbstractTaxol® (generic name Paclitaxel) is a chemotherapeutic drug, effective against head, neck, breast, lung, bladder, ovary, and cervix cancers. Rising demands in chemotherapy and limited supply of natural taxol have ultimately increased the cost of the drug. Semi synthesis using taxol precursors is not able to meet the global supply and has intensified the need to find alternative ways of taxol production. In the present study...

A simple, efficient and rapid screening technique for differentiating nitrile hydratase and nitrilase producing bacteria
Publication date: Available online 15 November 2019Source: Biotechnology ReportsAuthor(s): Ruchi Sahu, Anil Kumar Meghavarnam, Savitha JanakiramanAbstractNitrile hydrolyzing enzymes catalyze the hydration of nitrile compounds to corresponding amides and acids. Bacteria, isolated from soil samples were screened for nitrile hydrolyzing enzymes by simple dye based 96 well plate and nesslerization method. Bromothymol blue was used as an indicator for the detection of amides and acids based on colour change of the indicator dye from blue to dark green or yellow. The screening assay also differentiates between nitrile hydratase ...
