Detection of bovine viral diarrhoea virus nucleic acid, but not infectious virus, in bovine serum used for human vaccine manufacture
Publication date: Available online 23 June 2018Source: BiologicalsAuthor(s): Majid Laassri, Edward T. Mee, Sarah M. Connaughton, Hasmik Manukyan, Marion Gruber, Carmen Rodriguez-Hernandez, Philip D. Minor, Silke Schepelmann, Konstantin Chumakov, David J. WoodAbstractBovine viral diarrhoea virus (BVDV) is a cattle pathogen that has previously been reported to be present in bovine raw materials used in the manufacture of biological products for human use. Seven lots of trivalent measles, mumps and rubella (MMR) vaccine and 1 lot of measles vaccine from the same manufacturer, together with 17 lots of foetal bovine serum (FBS)...

Establishment of the first International Standard for human anti-typhoid capsular Vi polysaccharide IgG
Publication date: Available online 23 June 2018Source: BiologicalsAuthor(s): Sjoerd Rijpkema, Jason Hockley, Alastair Logan, Peter Rigsby, Eleanor Atkinson, Celina Jin, Haoyu Liang, Novilia S. Bachtiar, Jae Seung Yang, Akshay Goel, Venkatesan Ramasamy, Marcela F. Pasetti, anti-Vi IgG working groupAbstractVi capsular polysaccharide (Vi) conjugate vaccines prevent Typhoid in infants and young children. Several Vi-conjugate vaccines are being developed and an International Standard (IS) for human anti-Vi IgG is required to compare their immunogenicity. The suitability of 16/138 as an IS for human anti-Vi IgG was assessed alon...

Development of a relative potency test using ELISA for human rabies vaccines
Publication date: Available online 30 June 2018Source: BiologicalsAuthor(s): Zejun Wang, Yan Sun, Xianfu Wu, Darin S. Carroll, Wenli Lv, Lei You, Yaqi Ji, Jinrong Shi, Jiaxin Yan, Gelin Xu, Shengli MengAbstractThe NIH potency test for human rabies vaccines has disadvantages for use, especially in developing countries where rabies is endemic and prophylaxis needs ample, rapid, and reliable vaccine supplies. In China, 60–75 million doses of human rabies vaccines are administered each year. Vaccine quality control is of paramount importance, as is the release of potency-validated vaccines. We intended to design an alternati...

Evaluation of toll-like receptor 4 expression in human bone marrow mesenchymal stem cells by lipopolysaccharides from Shigella
In this study, we investigate the effects of various concentrations of LPS from different Shigella strains on TLR4 expression in human bone marrow (hBM)-MSCs. At the mRNA level, we have found that untreated hBM-MSCs (control) did not express TLR4 compared to the experimental groups. Cells treated with LPS from Shigella flexneri had the highest expression of TLR4, whereas cells treated with LPS from Shigella sonnei had the lowest expression. We observed that LPSs had a dose-dependent effect on TLR4 expression in all of the treatment groups. ELISA findings for interleukin-6 secretion have confirmed mRNA expression results fo...
