Development of a highly sensitive PCR/DNA chip method to detect mycoplasmas in a veterinary modified live vaccine
Publication date: July 2018Source: Biologicals, Volume 54Author(s): Sylvie Mbelo, Virginie Gay, Stephanie Blanchard, Eric Abachin, Stephanie Falque, Jacques Lechenet, Hervé Poulet, Blandine de Saint-VisAbstractMycoplasmas are potential contaminants that introduce undesirable changes in mammalian cell cultures. They frequently contaminate cell substrates and other starting materials used for manufacturing cell-derived biologics, such as vaccines and pharmaceutical products. Mycoplasma purity testing of live vaccines, active ingredients, raw material, and seed lots is required during vaccine production. Previously, testing ...

Publication date: July 2018Source: Biologicals, Volume 54Author(s): Roghaieh Didekhani, Mahmoud Reza Sohrabi, Ehsan Seyedjafari, Masoud Soleimani, Hana Hanaee-AhvazAbstractIn bone tissue engineering, bioceramics are of the most widely used materials for treatment of bone defects clinically. The composites of bioceramic/polymer fibrous scaffolds have been designed and developed to fulfill the mechanical and biological requirements of the damaged tissue. In the present study, oyster shell (OS) as a bioceramic in combination with the biodegradable and biocompatible poly (l-lactide) has been used to prepare a new tissue-engine...

Coculture of conjunctiva derived mesenchymal stem cells (CJMSCs) and corneal epithelial cells to reconstruct the corneal epithelium
This study aimed to find an improved and convenient system for the corneal epithelial differentiation of conjunctiva derived mesenchymal stem cells (CJMSCs). Thus, the cells were used to reconstruct corneal epithelial cells. Obtained by flow cytometry data, 51.9% of isolated CJMSCs were immune reactive for SSEA4+ antibody which are more potent to differentiate into corneal epithelial cells. A differential medium in a single culture plate was applied and compared to a Coculture with a SHEM medium and a Coculture with a commercial medium. It was found that CJMSCs can be induced to corneal epithelial cells through in vitro co...

Development of an anti-EPO antibody detection kit based on lab-on-a-chip and bridging antibody technologies
Publication date: July 2018Source: Biologicals, Volume 54Author(s): Jin-Gyo Oh, Jihyun Seong, Sunmi Han, Tae-Hwe HeoAbstractImmunogenicity is a major concern in the use of biological drugs. In particular, antibody-mediated pure red cell aplasia (PRCA) is a rare condition that is caused by administration of recombinant erythropoietin. There are numerous assay platforms for detect EPO anti-drug antibody (ADA), and most have appropriate assay sensitivity, but in need of improvement in terms of assay turnaround time and user accessibility. Here, the new method was developed based on lab-on-a-chip technology and bridging ELISA....

Evaluation of toll-like receptor 4 expression in human bone marrow mesenchymal stem cells by lipopolysaccharides from Shigella
In this study, we investigate the effects of various concentrations of LPS from different Shigella strains on TLR4 expression in human bone marrow (hBM)-MSCs. At the mRNA level, we have found that untreated hBM-MSCs (control) did not express TLR4 compared to the experimental groups. Cells treated with LPS from Shigella flexneri had the highest expression of TLR4, whereas cells treated with LPS from Shigella sonnei had the lowest expression. We observed that LPSs had a dose-dependent effect on TLR4 expression in all of the treatment groups. ELISA findings for interleukin-6 secretion have confirmed mRNA expression results fo...

Distribution and absence of generalized lesions in mice following single dose intramuscular inoculation of the vaccine candidate MVA-MERS-S
Publication date: July 2018Source: Biologicals, Volume 54Author(s): Martin C. Langenmayer, Anna-Theresa Lülf-Averhoff, Silvia Adam-Neumair, Robert Fux, Gerd Sutter, Asisa VolzAbstractModified Vaccinia Virus Ankara (MVA) is a highly attenuated and replication-deficient virus serving as vaccine against infectious diseases. Here, we assessed the in vivo distribution of a recombinant MVA candidate vaccine against the Middle Eastern Respiratory Syndrome (MVA-MERS-S) in mice. Intramuscularly inoculated mice were necropsied at different time points and examined by histology, immunohistochemistry and real-time PCR. We detected in...

Therapeutic monoclonal antibody N-glycosylation – Structure, function and therapeutic potential
Publication date: March 2018Source: Biologicals, Volume 52Author(s): Florian Cymer, Hermann Beck, Adelheid Rohde, Dietmar ReuschAbstractTherapeutic antibodies (IgG-type) contain several post-translational modifications (PTMs) whereby introducing a large heterogeneity, both structural and functional, into this class of therapeutics. Of these modifications, glycosylation in the fragment crystallizable (Fc) region is the most heterogeneous PTM, which can affect the stability of the molecule and interactions with Fc-receptors in vivo. Hence, the glycoform distribution can affect the mode of action and have implications for bio...
