β2‐adrenoreceptor medications and risk of Parkinson disease
We examined whether confounding by clinical indication for these medications, that is, tremor and smoking‐r elated pulmonary conditions, explained these associations.MethodsIn a large, population ‐based case–control study of United States Medicare beneficiaries in 2009 with diagnosis codes, procedure codes, and prescription data (48,295 incident PD cases, 52,324 controls), we examined the risk of PD in relation to use of selected β antagonists (propranolol, carvedilol, metoprolol), the β2 agonist salbutamol, and other medications used for the same clinical indications (primidone, inhaled corticosteroids). We adjust...

Neuroimaging and clinical outcomes of oral anticoagulant –associated intracerebral hemorrhage
ObjectiveWhether intracerebral hemorrhage (ICH) associated with non –vitamin K antagonist oral anticoagulants (NOAC‐ICH) has a better outcome compared to ICH associated with vitamin K antagonists (VKA‐ICH) is uncertain.MethodsWe performed a systematic review and individual patient data meta ‐analysis of cohort studies comparing clinical and radiological outcomes between NOAC‐ICH and VKA‐ICH patients. The primary outcome measure was 30‐day all‐cause mortality. All outcomes were assessed in multivariate regression analyses adjusted for age, sex, ICH location, and intraventricul ar hemorrhage extension.Results...

Development of a cerebrovascular magnetic resonance imaging biomarker for cognitive aging
ObjectiveRecent availability of amyloid and tau positron emission tomography (PET) has provided us with a unique opportunity to measure the association of systemic vascular health with brain health after accounting for the impact of Alzheimer disease (AD) pathologies. We wanted to quantify early cerebrovascular health –related magnetic resonance imaging brain measures (structure, perfusion, microstructural integrity) and evaluate their utility as a biomarker for cerebrovascular health.MethodsWe used 2 independent samples (discovery, n  = 390; validation, n = 1,035) of individuals, aged ≥ 60 years, along the...

Outcome of cell suspension allografts in a patient with Huntington's disease
For patients with incurable neurodegenerative disorders such as Huntington's (HD) and Parkinson's disease, cell transplantation has been explored as a potential treatment option. Here, we present the first clinicopathological study of a patient with HD in receipt of cell ‐suspension striatal allografts who took part in the NEST‐UK multicenter clinical transplantation trial. Using various immunohistochemical techniques, we found a discrepancy in the survival of grafted projection neurons with respect to grafted interneurons as well as major ongoing inflammatory a nd immune responses to the grafted tissue with evidence o...

Disease progression and outcome measures in spinobulbar muscular atrophy
In this study, we prospectively investigated disease progression over 18 months in 29 patients with genetically confirmed SBMA, using quantitative outcome measures, including Dixon magnetic resonance imaging (MRI).MethodsWe used MRI to assess changes in muscle fat content and stationary dynamometry to assess changes in muscle strength. Disease progression was also investigated with the SBMA functional rating scale, bulbar rating scale, 6 ‐minute walk test, and blood samples, among others.ResultsMean muscle fat content, muscle strength in knee extensors, handgrip strength, walking distance, and creatinine levels changed s...

NBEA: Developmental disease gene with early generalized epilepsy phenotypes
NBEA is a candidate gene for autism, and de novo variants have been reported in neurodevelopmental disease (NDD) cohorts. However,NBEA has not been rigorously evaluated as a disease gene, and associated phenotypes have not been delineated. We identified 24 de novoNBEA variants in patients with NDD, establishingNBEA as an NDD gene. Most patients had epilepsy with onset in the first few years of life, often characterized by generalized seizure types, including myoclonic and atonic seizures. Our data show a broader phenotypic spectrum than previously described, including a myoclonic ‐astatic epilepsy–like phenotype in a s...

Relative neuron loss in hippocampal sclerosis of aging and Alzheimer's disease
ObjectiveTo characterize the pattern of neuron loss in hippocampal sclerosis of aging (HS ‐Aging) and age‐related diseases and to evaluate its contribution to cognitive impairment in the elderly.MethodsParticipants (n  = 1,361) came from longitudinal observational studies of aging at the Knight Alzheimer Disease Research Center, Washington University (St. Louis, MO). Relative neuron loss in the hippocampus of HS‐Aging was measured using unbiased stereological methods. Transactive response DNA‐binding prot ein of 43 kDa (TDP‐43) proteinopathy, a putative marker of HS‐Aging, was assessed. Clinical and cogniti...
