Perspectives for the Comet Assay in Assessing Genotoxicity - A Review
The employment of comet assay has gained wide importance in various areas that focus on human bio-monitoring, genetic toxicology, molecular epidemiology and ecogenotoxicity. Comet assay has become among the standard methods for evaluating genotoxicity by the determination of DNA damage caused by mutagens, and this assay has been extensively applied by the researchers for the past 20 years. This review discusses comet assay, as well as its application in various fields. (Source: Current Pharmacogenomics and Personalized Medicine)

Genetic Variations in Cytochrome P450 2C9 and 2C19: A Review
DNA polymorphisms have an important role in the genes encoding. A number of cytochrome P450 (CYP) enzymes are leading to drug metabolism variation in different subjects. CYP2C9 and CYP2C19 metabolize many clinically important drugs. Inter-individual variability in drug metabolism may change drug reactions and variability in drug efficiency. Cytochrome P450 (CYPs) polymorphisms may show an important effect on drug metabolism, toxicity and hepatotoxicity. The most common clinical effects are associated with CYP2C19 and CYP2C9 mutants. Differences in genetic polymorphism of these cytochromic enzymes in inter-individual still ...

Practical Challenges and Translational Issues in Pharmacogenomics and Personalized Medicine from 2010 Onwards
Conclusion: Costs in the drug discovery process and the implementation of personalized medical approaches are the main barriers to the translation of pharmacogenomics from laboratory to clinic. The lack of a unified system as well as the requirement for expert analysis of data is additional barriers. (Source: Current Pharmacogenomics and Personalized Medicine)

Tailoring of Medicinal Plant Treatments to the Individual Characteristics of each Patient with Precison Medicine Approach
Drugs are generally tested on a large population of people via phase studies and the average response is reported in a sort of evidence-based medicine. Screening of genetic variations is necessary in understanding the underlying mechanisms of individual drug responses and to find out more targeted and unique therapeutic strategies focusing on this individual variations. The “-omics” (genomics/proteomics/metabolomics) technology can enable researchers to further understand the causative mechanisms/ biomarkers. Furthermore, they might be used to assess optimized drug efficacy and safety. The most common indications for t...

Epigenetics and Adult Acute Myeloid Leukemia
Acute myeloid leukemia (AML) is the most common leukemia diagnosed in adults and curative treatments remain currently a great challenge. AML occur in about 60% of patients over 60 years with unsatisfactory survival results, which have not improved significantly over the last two decades, in spite of improved supportive care. Global genomic instability is the hallmark of the disease, leading to chromosomal/molecular alterations, proliferative advantage and clonal heterogeneity. Epigenetic deregulation of gene expression is considered as a consequence and/or the initiating cause of this instability and DNA methylation is one...

Beneficial Effects of Dietary Supplements Against Mitochondrial Dysfunction and Apoptosis in Neurodegenerative Diseases
Conclusion: In conclusion, we made an attempt to give an overview on the PTMs of mitochondrial proteins and their importance in understanding the role of mitochondrial dysfunction and apoptosis in neurodegenerative diseases. Furthermore, we described the PTMs (oxidation, nitration, phosphorylation, etc.) of mitochondrial proteins that may play an important role in mitochondrial dysfunction and promote neuro-degeneration. We also concluded that ingesting healthy diets combined with physical exercise may restore normal mitochondrial functions (Fig. 1). Furthermore, a better understanding of the mitochondrial dynamics as well...

Biological Basis of Ageing, Age-related Diseases and Interventions
Background: The survival of all biological systems depends on a series of complex pathways of maintenance and repair systems, which are the crucial components of the homeodynamic space of an individual. A progressive failure of maintenance and the consequent shrinkage of the homeodynamic space are the fundamental basis of ageing, age-related diseases and eventual death. Mechanisms: Although longevity assurance genes do affect the essential lifespan of a species, there are no gerontogenes with the specific function to cause ageing and to limit the lifespan of an individual. Approaches: Age-related diseases and other healt...

Antidepressant-Like Activity of Fish Oil, with and Without Fluoxetine, in Two Behavioral Despair Paradigms in Experimental Mice
Conclusion: FO displayed anti-depressant - like action suggested by the FST and TST. Combined treatment with fluoxetine and FO did not significantly affect the performance of mice on these two tests. This suggest FO has direct bearing on the present reported behavioral paradigm of despair and helplessness. Future studies to explore molecular the molecular mechanisms behind the presently observed behavioral effect of FO in this animal model. (Source: Current Pharmacogenomics and Personalized Medicine)
