Fixable Stain for Flow Cytometric Evaluation of Stallion Sperm Viability
Sperm membrane integrity is an important component of stallion sperm quality and fertility potential [1] that is typically assessed at the region of the sperm head and considered a measurement of viable or live sperm [2]. Flow cytometric assessment of viability involves the collection and transport of live sperm to a laboratory for evaluation, with assays run on unfixed sperm. A double-stain assay uses SYBR-14, a penetrating nucleic acid green stain, and propidium iodide (PI), a red stain that labels the nucleic acid of membrane-compromised sperm [3].
