Protective effect of extracellular polysaccharides from Grifola frondosa mycelium on CCl4-injured liver in vitro

As the study showed, EXO-GFP was mainly consisted of α-pyranoside glucans. Four monosaccharides including rhamnose, arabinose, xylose, glucose were contained, with the molar ratio at 0.028:0.031:0.030:8.98, respectively. The protective role of EXO-GFP was researched in cell cycle, detection of reactive oxygen species (ROS) by fluorescence staining, detection of cytochrome P4502E1 (CYP2E1) and tumor necrosis factor alpha (TNF-α) by western blot. Compared with the cell of model group induced by CCl4, AST, ALT activity of EXO-GFP treated group decreased significantly, and the blockage of S phase induced by CCl4 was reduced significantly. EXO-GFP could decrease the over-active of CYP2E1, TNF-α and ROS fluorescence induced by CCL4. These results indicated that significant protective effect of EXO-GFP on hepatic cell (L-02 cell line) injury induced by CCl4 in vivo. Graphical abstract
